Make a circle with a thick stroke and no fill. Duplicate it and offset the location, set the blend mode to Erase. Now put the appropriately sized letter underneath those two circles with a cropped copy on top of the stuff. Group the four of them to ease moving it around.

I have never mastered color management, period, so I cannot help with that.

I was able to recreate your design using parts of your design. But I was not able to create that circle.

 

I would really appreciate if you could show me in more details how you created that beautiful circle.
