Have you opened the PNG file in your (which?) Affinity app? Did you select it with the Move tool?

If so, in whatever asset sub-category you want to add it to, if you click on the 3 line 'burger' menu next to that subcategory, "Add from Selection" should not be greyed out.

If it is, can you provide a screenshot showing the selected item & what the menu shows?

If you are using the ‘Import Assets…’ command/option from within the Assets window it will only let you import .afassets files, not other general file types.
But you can use ‘Drag & Drop’ to import the entire folder of images you want or you can use Drag & Drop with individual images from said folder to import them.

Create your new category and then Drag & Drop the folder or selected items from within it into this new category in the Assets window.

From what you show, I think @markw is right -- you are trying to use Import Assets for this, which only works with *.afassets files (which are collections of assets which can be created via the Export Assets in the same menu). To import individual files like PNG's, you need to do that like I mentioned above, by opening the file, selecting it, & using the Add from Selection item in one of you asset subcategories.

Or the method suggested by @markw of drag/dropping from File Explorer or Finder; just not by using Import Assets.

What you are showing is a locked layer. This does not seem to have anything to do with assets.
