As you can see in the attached file: 1) the opacity of the square is 100%, the opacity of the stroke is set to 100% too. 2) The value 60% does not exist in the menu Thanks for your help. Quote

Your second appears to show a different document with a single pixel layer, and a red brush. Did you upload the correct screenshots? It would help to have a screenshot of the complete application window, with the Brush Tool active, and the Color panel open.
