How to place an object into another object with specific measures?

Let's say I have two squares. a) Big one. b) Small one.

 

I want to put the Small One into the Big One in the way that the distance between the right side of the small one and the right side of the big one becomes 6 cm.

 

How can I do this?

Activate grid, or guides, or create a helper object of 6cm.

1) How can I use Grid feature for this purpose? (So It has to show the measurement or snap to the grid on the exact measurement.)

You can create a rectangular shape and use the transform panel to give it exactly 6cm width / height. 
Then, with snapping active, position it to the big object so that snapping visually kicks in. 
finally, you can move you smaller object to snap to the helper object with exactly 6cm distance.
