I have the doc "A Guide to installing creative resources downloaded from the Affinity Store." The Assets Studio is mentioned, and it tells me to import assets through the Assets Studio, but it doesn't tell me how to get those assets onto the iPad in the first place. Does the Assets Studio make a connection to my account on the Affinity Store and let me choose which to import?

When you tap import from the Asset Studio, it opens into Files app. You can then navigate to location when you have stored the .assets file to select and import it. If you have issues, try storing the file on iCloud and import from there.

At the moment, all my assets are on my iMac. Should I connect the iPad to the iMac and drag them on to the iPad or, in the case of Frankentoon, for example, go back to their store on my iPad and try to re-download them?

Transferring across from Mac will work fine. Some people have had issues importing assets on current release but this has been fixed in beta. Also may need to be on iCloud. Seems to effect different models in different ways.
