Adjusting the Sharpen Filter

I tried sharpening the eyes of a rendered 3D character. I could not see much difference. I'm just getting started using Affinity Photo, as of today. I've been using Photoshop. What would be the settings if I wanted a 100% sharpening? Also, is there a downloadable text tutorial, or information about using Photo? I've read books on Photoshop. So, I know how to use Photoshop. It's just that I'm a bit confused with Photo.

For example, if I want to sharpen a photo, do I just create a new layer and apply the filter? Can I duplicate the original photo layer and sharpen that? Sorry to sound so ignorant. Just getting started. I know how to do these things easily in Photoshop. I'm not sure how to apply a sharpening filter to the extent that would please me.
