Export with transparent layer

Does exporting an imported .svg file as '.png with transparency', the same as unticking the white '(Rectangle)' tick box in the right-hand column under 'Layers'?

If i understood you question correctly, no. Hiding the "white" rectangle just hides that object (no matter if it fills the canvas or not) from view. To make the background transparent go to File ▸ Document Setup..., Colour section and tick Transparent background.
