Using Affinity Designer is there a simple keyboard shortcut that can switch the eraser tool between remove pixels and adding pixels back in?

No, but if you create a mask layer in the Pixel Persona, you can paint on the mask with black paint to delete the image, then press X to swap to white and paint the pixels back in.

 

Nice and non-destructive too.

Adding very slightly to what @toltec said, note that the Erase Brush Tool only has one 'mode' -- it erases pixels regardless of the brush color.
