Current b/w drawing (JPG) has only gray scale palette. Need to add colors

Which Affinity app are you using? In Designer, choose 'Document Setup...' from the File menu and then go to the Colour tab in the dialog and change the colour format (e.g. from 'Grey/8' to 'RGB/8'). In Photo, the Colour Format option is on the Document menu.

 

If the drawing is identified in the Layers tab as an Image layer, it can only be transformed globally (e.g. rotating the entire layer). To work at the pixel level you need to right-click the layer (in the Layers tab) and choose 'Rasterize' to turn it into a Pixel layer.

 

In the Pixel Persona in either app you can use the Selection Brush Tool and/or other selection tools to define the closed space that you want to fill with a colour. You can then use the Flood Fill Tool or the Paint Brush Tool to apply your choice of colour.
