Will Designer run on old Macbook?

Yes, I have been running AD on my 2009 MBP. Yes I do have Safari freezing now and again and the spinning beachball. so for a seven year old machine I'm not ready to chuck this machine out just yet.

Affinity Designer requires a 64 bit CPU. If the MacBook has a Core Duo processor it is 32 bit. A Core 2 Duo is 64 bit. See Apple's list of CPU types here.
