If you mean new document, that's in the Preset you choose, or in the File > New dialog, as Hens has mentioned. It's also in the Assistand Manager, in the Develop section, for Opening RAW images and Developing them. There you can choose 32-bit or 16-bit as the result of the Develop operation. I am curious, though, why you want everything in 32-bit. It's a very different working environment, and James Ritson has recommended several times not using it unless you're using one of the very specialized workflows that need it.
