I have edited & converted an image to black & white ready for printing.

When setting the dialogue using the above printer, does ticking the black & white box alter in any way the image ? or should I leave it "unticked" ?

These are just guesses based on my experience with other printers.

I think the B&W box means only the Black Ink will be used. No colour ink will be used. If it is unticked then perhaps colour ink will be used.

Mac Pro (Late 2013) Mac OS 12.7.6
Affinity Designer 2.6.0 | Affinity Photo 2.6.0 | Affinity Publisher 2.6.0 | Beta versions as they appear.

I have never mastered color management, period, so I cannot help with that.

No, since canon already tells you for this option setting that ...

During Black and White Photo Print, inks other than black ink may be used as well.

It is possible to perform color/tint adjustments in the Pro-100 driver.  If Black and White is turned off, then on the "Main" tab of the driver dialog you select "Manual" for Color/Intensity then "Set" you get a panel where you can adjust CMY values for the print as well as Brightness, Intensity and Contrast.

If you turn on Black and White that panel becomes different, oriented towards monochrome printing.
