My question is relatively simple, but I'm new to Publisher and I'm not finding any tutorials.
Is it possible to create column guides with different widths? Something like the image below

Make your text frame and use the text frame palette to adjust amount of columns, gutter and column widths. You can also just drag gutters to desired positions once you have enough columns.

Try using Immediate Layers and Children in order to snap to the objects from a master page.

I have never mastered color management, period, so I cannot help with that.
