As you can see below, I am unable to change the alignment of a single line, made with the pen tool, to outside or inside.

It is possible with the other three figures, these are also made with the pen tool?

Is there any way to get this fixed for a single line?

Answer the question: Where is the outside or inside of that line? 

There is only the line, it has no inside or outside.

I have never mastered color management, period, so I cannot help with that.

Sure, but it could be used to align a left/right line stroke without having to change its location.

My question was actually also related to a question I had previously asked:
