Center Red and Green Snap Guides Missing

Make sure "Snapping" is on (the magnet at the top) and it should work.

In addition to having Snapping enabled, you need to have the right Snapping options set. Do you have Snap to Spreads and its suboption Snap to Spread Midpoints enabled?

No, Lock Guides does not affect it. I'm not sure what's going on.

I know that this is an old thread, but I found it because I ran into the same problem in Publisher 1.10.5.  From my experiments, it seems that getting the green and red center alignment indicators is controlled by turning on "Snap to object bounding boxes" as well as the subordinate "Include bounding box mid points".  Strangely, I wasn't able to get the red and green lines to appear until I unchecked ALL of the other snapping boxes (except for the "Enable snapping" one at the top.  After I got this working, I was then able to check the "Snap to spread" and "Snap to margin" options, as well as the subordinate "Include spread mid points" and "Include margin midpoints", which were previously checked before I unchecked everything.  I'm guessing that it's possible that the order in which the snapping boxes are checked might affect the order in which the criteria for snapping are evaluated, since it appears that the "Screen tolerance" value might prevent the red and green lines from appearing if there other higher-priority snapping candidates are activated before the dragged object is able to be center-aligned.
