Yes it's possible.

1. Create a new file

2. Before you create anything on the art board, go to stroke panel.

3. Tick 'scale with object'

4. Go to 'Edit' menu

5. Select 'Defaults' and then select 'Save'

These settings become default and the next time you create a new document these will be effected.

Remember that you can also use same process to edit and save settings for default font, font size, stroke and fill properties etc... You can effectively remove the annoying default fill colour in order to remain with the stroke only if you so wish.

Assuming that one is currently using the later versions (1.7 or 1.8), they can also utilise the new template feature and save the settings as a new template.
