Does Affinity Photo use fonts from the Mac's Library (System or User?). How and where do you add new fonts? Will Affinity access Mac's Font Book app. ?

Yes that's right, AP use Mac's Font Book app

You can also use User/Library/Font. That's the folder Font Book uses

Like most apps, it uses fonts from three Library folders. Their file paths are as follows:

  • System/Library/Fonts
  • Library/Fonts
  • Users/<user>/Library/Fonts (usually abbreviated as ~/Library/Fonts)

Fonts in the first two folders are available to all users; those in the last one only to the current user. System fonts cannot (easily & safely) be removed or disabled.

Font Book is the default Mac OS font manager. The Mac Affinity apps support its features, including in the Character Studio panel allowing you to filter the panel's popup font list by a collection or "Smart Collection" created in Font Book. (Note: do not name a Font Book collection "Favorites" because it will conflict with the Affinity Favorites list, causing list selections to be off by one.)

Assuming you are using  Font Book as your font manager, you can use it to install, enable & disable, remove, download, validate fonts, & more. To learn about its options, I suggest opening that app, choosing "Font Book Help" from the Help menu & browsing through its topic list.
