This is not relevant here. The problem is that if you add a 1 px wide inner outline, everything will be covered except for the character.

I can confirm it. Extremely strange!
As if the character had been cut out.

I have no clue what the problem could be, I have taken the picture to APhoto and even there the picture is displayed in such a way that the character is shown with a black background cut out.

I think this font isn't free nor free for personal use. The font family is listed on myfonts https://www.myfonts.com/de/collections/filson-pro-font-mostardesign and some other font stocks

To remove the areas, switch to the Designer Persona.
Select the letter and the area to be removed. To do this, hold down the Shift key. Now click on the Substract geometry symbol in the toolbar.
After removing the O in Integration, a dark grey curve is still visible; this can be deleted.

I have done the work for you once. You can see what I have done in the history save with the document.

Setze den Ebenen-Mischmodus der weißen Linien auf Radieren. Durch Änderung der Deckkraft ändert sich die Farbmischung und somit bekommst Du einen anderen Farbton im Mischbereich.

Ja, kann auch nicht denn mit 53 KB ist die Datei definitiv nicht wiederherstellbar. Eine von mir erstellte Testdatei einer Seite A4 mit Fülltext hat schon 193 kb.

Möglicherweise ist eine Autosave Datei vorhanden. In das Verzeichniss kommst Du mit

MSI/EXE Installation        Windows-Taste+R > %appdata%\affinity\publisher\2.0\autosave\ 
MSIX Installation       Windows-Taste+R > %USERPROFILE%\.affinity\publisher\2.0\autosave\

Die .autosave Datei solltest Du an einen anderen Ort kopieren und duplizieren. Das Duplikat dann umbenennen in z. B. masterarbeit.apub oder einem anderen, dir genehmen Namen.

Hast Du viele Dateien im Dokument verlinkt? Wenn ja, könnte es helfen, den oder die Speicherorte der Dateien vorrübergehend umzubenennen.

You can preview the saved png in Windows Explorer. All you have to do is to activate the details view.

If you don't see a white background in your cropped png file, you've done everything right.

And, you don't asking to much. Nobody does know all.

The handwritten text should have a strong dark colour. You can then select the Blend options in the Studio tab Layers on the right-hand side of the cogwheel.
In the Source Layer Ranges section, set a handle point to the middle, select the right handle point and drag it down and towards the centre. The background should now slowly become invisible. Now you can crop the layer and execute the Rasterise and Trim command via Menu > Layer.
Now export as png or pdf.
