I suggest watching Affinity Photo - Layer Concepts for an in depth demonstration of how this works, but very briefly:
Under the layer -- affects only layers below
On the layer -- a "child" layer that affects only the parent layer
Above the layer -- affects all layers below

Under – adj layer affects only layers below it, so if it is lowest it does not do anything
On – affect only current layer
Above – affects all layers below
